DNA Profiling (DNA Fingerprinting) | A laboratory technique to assist in the identification of individuals by their respective DNA profiles. It gained its name because the DNA sequence of each person is different, and hence, just as each of us is unique with respect to the pattern of our fingerprints, we can be identified from our DNA. Can be used for parental testing and criminal investigation. |
Restriction Fragment Length Polymorphism (RFLP) | A technique that exploits variations in homologous DNA sequences. It refers to a difference between samples of homologous DNA molecules that come from differing location of restriction sites. In RFLP analysis, the DNA sample is broken into pieces (digested) by restriction enzymes and the resulting restriction fragments are separated according to their lengths by gel electrophoresis. |
Restriction Enzymes | Also called restriction endonucleases, these are molecular scissors that cut DNA at a specific site with a specific nucleotide sequence (restriction site) of 4 to 8 bases in length. For a given length of DNA, if the restriction site occurs more than once, the restriction enzyme will cut each of those sites, thus fragmenting the DNA. |

Ethidium Bromide | An intercalcating agent commonly used as a flourescent tag (nucleic acid stain) in molecular biology laboratories for techniques such as agarose gel electrophoresis. It is commonly abbreviated as "EtBR". When exposed to ultraviolet light, it will flouresce with an orange color, intensifying almost 20-fold after binding to DNA. Ethidium bromide may be carcinogenic. |
