What is the simple staining and what was the purpose of this exercise ?
simple staining involve only one stain, and the purpose is to identify the parts of the bacteria
1-Bacillus subtilis = shape, long, oval
2-Staphylococcus Epidermis = shape oval, circular balls
why would one make sure that the commercially available glass slide does not have a layer of grease before preparing the smear?
because you want whatever you put on the slide to stick so you can stain it. If there is a grease, your specimen wont stick to the slide when you fix it and as you apply your stain you will wash off your specimen
what would happen if you had skipped the heat fixing step and continued wit the staining procedure?
The bacterial cells on the slide would shift when the agents are being rinsed off which would mean the bacteria would go down the drain
what is the reason for sterilizing the whole wire and part pf the handle of the inoculating loop before and after each transfer?
to insurance that the bacteria is free of contamination from other microorganisms that could be surround it
why is knowing the bacterial shape and arrangement important?
because enables scientists to determine what type of bacteria it is and what methods need to be used to control the growth
