1- no enough bacteria to spread out everywhere to thin when distributed to the second and third section
2- If after heating the loop between spreading section one did not wait for the loop to cool the bacteria will be kill
3- the agar was not proper distributed in the plate
how would you determine which colonies might be contaminants on a streak plate ?
if you see a color that you did not expected, or does not match. Also, if you see a different shape of bacteria that you cannot identify
