Capsule: How is the basis of a negative stain different from the other staining protocols that we performed so far in lab? This is not asking just about the capsule stain, which is only one example of a negative stain.
Negative stains are often used for cells that are too delicate for heat fixing. This is why negative staining method is so different from some of the others we performed in class where heat was used as either a mordant or to fix the sample.
Capsule: (T/F) The capsule stain is the only multistep protocol that we performed that does not follow the standard four step differential process.

We usually use heat to fix the sample. For this particular stain we used acid alcohol.

In capsule staining, acid alcohol is used to chemically fix the cell to the slide. For the acid-fast staining exercise we used acid alcohol as a decolorizer

my particular sample had a dark purple background with hollow cells.
Capsule: You should be able to look at a slide and tell that it is a capsule stain versus other staining procedures that we did in lab.

-do not rinse off, pour off
-acts as mordant/fixes the cell

-put on slide before anything else. just one small drop
-mix with 1 loopful of bacteria and let air dry

-you wont rinse until the counter stain has been added
-do not allow water to run over top of slide (bottom only)
-bolt dry bottom and edges only
-start at 10x
-only use fine adjustment knob at 40x
-oil at 100x
