With a lab marker, label the bottom of each tryptic soy agar (TSA) plate with your initials, your lab section and seat number, the name of the organism to be inoculated, the temperature, and the date.
2. Remove a sterile loop from the bag.
3. Pick up the broth culture of bacteria with your free hand (the one not holding the inoculating loop) and mix well by gently flicking the bottom of the tube. Remove and hold the cap of the culture tube with the small fingers and palm of the hand holding the sterile inoculating loop. Do not lay the cap on the table, which could result in contamination of either the culture or working area. Holding the tube almost horizontally, flame the mouth of the tube by quickly passing it through the gas burner flame twice. Do not hold the tube in the flame.
4. Insert the sterile loop into the open culture tube (holding both almost horizontally) and remove a loopful of culture. Be careful not to touch the sides of the tube or the lip with the loop. Hold the loop steadily while proceeding with the next step (remember that the loop is loaded with millions of microorganisms).
5. Flame the mouth of the tube again, replace the cap, and return the tube to the tube rack.
6. Pick up the uninoculated Petri plate with your free hand and lift the lid slightly. Insert the "loaded" inoculating loop and touch the loop to the surface of the agar lightly. Slide the loop gently over the surface in a continuous motion, starting near the outer edge. and moving towards the center, covering one-fourth to one-third of the plate. Keep the streak lines close together without crossing back over a previously streaked region.
7. Dispose of loop and remove another sterile loop from the bag.
8. Cross over the streak pattern in the first area 3-4 times and continue streaking in a second, adjacent region.
9. Dispose of loop and remove another sterile loop from the bag.
10. Cross over the streak pattern in the second sector 3-4 times and continue streaking in this third, adjacent region.
11. Dispose of loop.
