Which diagnostic method uses phenotype to diagnose via a microscope as well as performance of various tests on the organism, morphology, immunological, or DNA? Which method uses PCR? Which is quickest and least expensive?
Morphology (look under microscope), PCR is DNA method, Immunological - ELISA quick and easy
Which morphology uses a fluorescent antibody technique to reveal antigens?
Microscopy with Fluorescent Antibody Techniques (direct)
When discussing slides 5 and 6, Dr Tan talks about "grabbing antibodies from your lab freezer". How were these antibodies made?
First, inject animal with specific organism so primary antibodies are created. Then store these in your lab freezer.
Why does the secondary antibody used in indirect fluorescent antibody techniques need to be from a different animal than the primary antibody?
Secondary has to be a different animal b/c if you use the same animal, it will not see it as foreign ... therefore, it wouldn't cause a reaction to create antibodies.
Put these steps for direct fluorescent antibody technique in proper order:
A. Heat fix the slide
B. Rinse the slide
C. Swab organism and put on slide
D. Look at the slide under fluorescent microscope
E. Instead of a stain apply fluorescent antibodies you got from your lab freezer
c, a, e, b, d
Each animal has its own specific Fc domain that is universal for each species. T/F
True
With indirect fluorescent antibody technique, which antibody binds to the antigen, primary or secondary? What binds to the primary antibody?
Primary antibody binds directly to antigen. Secondary binds to the primary antibody.
You are at an in-person lab and have been asked to test a sample for Staphylococcus aureus. You perform an indirect fluorescent antibody test. The result is positive. When your lab TA checks your work, she lets you know the result should have been negative. What step did you most likely forget?
You most likely forgot to rinse off the slide. In a properly done positive test the secondary will only bind if the primary survives the slide being rinsed off.
What type of key uses traits to ID organisms that have undergone biochemical tests?
Dichotomous Key
Which test is more of a shotgun approach where you can do multiple biochemical tests at a time, PCR, FISH, or API? Which is run many times to multiply organisms?
API; PCR
In which test would the presence of a precipitation ring give a positive result? Why are these not used much any longer?
Precipitation reactions; Requires a lot of material
Agglutination reactions can be used to determine blood type. They involve particulate antigens or antibodies. The antibodies can attach to many blood types. The pattern of strings tells you the particular blood type. These tests do not need a lot of material. There are 4 mistakes in the statement above. Find each one and replace with correct info
"or" should be "and"
"many" should be "one"
"strings" should be "clumping"
"do not" shouldn't be there as they usually do need a lot of material
Which test uses gel electrophoresis, the ELISA or Western Blot? Which is used to make inexpensive pregnancy tests? Which would be used to detect HIV? Which uses enzymes?
Western Blot; ELISA; Western Blot; ELISA
Which ELISA tests for a specific antigen in the sample, direct or indirect? Which one is used in pregnancy tests?
Direct; Pregnancy tests are direct ELISA
Put these direct ELISA test steps in order (you may use options more than once if needed):
a. Rinse out tube
b. Add substrate
c. Add primary enzyme linked antibody
d. Add primary antibody
e. Add patient sample with antigen
d, a, e, a, c, a, b
The tube from your test above turns blue. What does this mean?
Substrate got cleaved into product so the enzyme-linked antibody in the test region of the strip bound to it ... which turned it blue
What is the advantage of using an indirect ELISA vs a direct ELISA?
Signal amplification
