Individual Dilution Factors
volume of sample diluted/ total volume diluent + sample volume. Ex: 1mL/9mL + 1mL(sample) = 1mL/10mL = 10^-1 IDF
Total Dilution Factor
multiply the individual dilution factors together. Ex: 10^-1, 10^-2 and 10^-3 then the TDF=(10^-1 x 10^-2 x 10^-3)= 10^-6 Note what when multiplying by powers of ten you add the exponents.
How to calculate cell concentrations using serial dilutions
[Cells] in original sample= # of cells from tube X/ (TDF tube X) x (volume tube X examined)
Ex: 100 cells/mL/10^-3= 1.0 x 10^2 cells/mL/10^-3= 1.0 x 10^2-(-3)= 1.0 x 10^2+3= 1.0 x 10^5 cells/mL
When dividing exponents...
the exponents in the denominator (bottom)can be subtracted from the exponents in the numerator (top)
When multiplying exponents...
the exponents can be added together
accuracy
is how close a measurement is to the true, correct or actual value.
Precision
is the repeatability of a measured method, but doesn't necessarily imply accuracy.
