what are the key stages where interruption is possible in HIV replication?
1. binding
2. fusion
3. reverse transcription
4. integration
5. maturation
what are Nucleoside reverse transcriptase inhibitors (NRTIs)?
- inhibit transcription of RNA to DNA
- OBLIGATE DNA CHAIN TERMINATOR
- stops new viral infection but not viruses that have already infected cells.
what are non-nucleoside reverse transcriptase inhibitors (NNRTIs)?
- distort reverse transcriptase enzyme so blocks conversion of RNA to DNA
what are fusion inhibitors
- prevent virus from initially entering the cell by inhibiting fusion
what are protease inhibitors (PIs)?
- prevent viral maturation so HIV not assembled and released
- bind to proteases and prevent correct cleavage of viral proteins
how does HIV establish drug resistance to NRTIs?
steric hindrance = change in shape so drug can no longer bind
how does HIV establish drug resistance to NNRTIs?
reduce interactions between reverse transcriptase and drug via steric hindrance (drug cant bind properly so cant inhibit reverse transcriptase)
